Base Pair Biotechnologies, Inc. services
Assay Development Services
While aptamers and antibodies are used in many of the same applications because of their selective binding capabilities, aptamers and antibodies are structurally very different. Ideal buffer formulations, assay conditions, and optimal reagent concentrations can vary significantly between aptamer-based and traditional antibody-based assays. Base Pair offers aptamer-based assay development services for a wide range of applications, including lateral flow assays (LFA), ELISA-like assays (ELASA), dot blots, western blots, and many others. Base Pair also offers advanced aptamer selection, characterization, and optimization techniques to improve real-world, application-specific performance.
Bioinformatics for Aptamer Selection
Aptamer discovery is a complex process. Following library enrichment against the desired target, the best aptamer candidates are identified from millions of potential sequences. Whether you are new to aptamer selection or simply short on manpower, Base Pair is here to help. Following next generation sequencing, up to 10 million sequences are evaluated using proprietary bioinformatics techniques. A FASTQ file containing sequence information from final aptamer pools or pools from several selection rounds is uploaded to a secure ftp site.

Custom Aptamer Discovery & Development Services
Because aptamer performance is optimal under selection conditions, we recommend that you consider a custom development project for any specialized assay or use conditions. Base Pair’s custom aptamer discovery and development service is highly customized to meet each customer’s unique needs. Here is a general overview of our aptamer development process, describing key features of each phase. As we work with you to understand your project, we will develop and share with you a detailed plan, aligned with your goals and objectives.
Aptamer Maturation
Aptamer discovery is a complex process. Customized libraries and selection techniques are employed to achieve selective binding to a target of interest under anticipated conditions. In some cases, selected aptamers may not quite achieve the degree of affinity or stability required for a specific end-use application. In addition, aptamer modification for immobilization or signal generation is often required. Following initial selection, a wide range of tools and techniques are used to evaluate and improve performance for DNA and RNA aptamers. Aptamer modification is often an iterative process. Base Pair’s in-house selection, sequencing, bioinformatics, and affinity determination capabilities make successful aptamer maturation possible. Whether your aptamer of interest was discovered at Base Pair or in another laboratory, Base Pair can help select the best modifications based on your individual needs and end-use applications.
