Pladienolide B is the major analogue of a family of macrocyclic lactones isolated from Streptomyces platensis. Pladienolide B is a highly potent splicing inhibitor of both hypoxia signals and cancer cell proliferation. Pladienolide B binds to the SF3B1 subunit blocking the spliceosome. Through this action, Pladienolide B stops the growth of cells proliferating by inhibiting mRNA splicing.
