Systemic lupus erythematosus (SLE) is a classic systemic autoimmune disorder characterized by impaired autoimmune tolerance and high concentration of autoantibodies targeting self-antigens, which lead to organ damage. This pathological process involves multiple cytokines. Notably the role of CD40L (CD154) has been extensively studied in SLE, where CD40L is markedly upregulated. In both human and murine models, sustained CD40L expression in lupus T cells can drive excessive activation of B cells, contributing to the pathogenesis of disease. Elevated soluble CD30 levels in SLE serum mark for high disease activity. IL-10, a key Th2 cytokine, is implicated in this autoimmune dysregulation. Additionally, IL-13 has been a significant focus in SLE research. Increases in pro-inflammatory cytokines such as IL-6, IL-12, IL-18, and TNF-α can initiate the inflammatory process of SLE. These cytokines are found elevated expression in SLE patients and correlated with disease activity. It is well-documented that interferon-α levels are elevated in the serum of patients with active SLE, and this cytokine is responsible for the formation of vesicular inclusions in the renal endothelial cells of these patients.
